Misao is a very energetic and cheerful girl, always seeing the good in people. She holds strong admiration for the other members of the Oniwabanshu, partcularly her dear Aoshi-sama. Unfortunately, she does have a short temper and is quick to lash out her annoyance on the people around her.
Misao is the granddaughter of the former okashira - leader - of the Oniwabanshu, a clan of ninjas based in Kyoto. As a child, she was in the care of the young okashira Shinomori Aoshi until Aoshi and his four most trusted men - Han'nya, Shikijo, Beshimi, and Hyottoko - left her behind in Kyoto with an elder member known as Okina. She was only eight but her memories of the five of them remained engraved in her mind and heart.
At sixteen, she began a series of trips in search of her beloved Aoshi-sama whenever she received the slightest news of his whereabouts. When she heard that Aoshi was in Tokyo, she went to search for him and runs into Himura Kenshin, who she doesn't realize is the legendary Hitokiri Battousai of the Bakamatsu on the way back to Kyoto. They ended up travelling to Kyoto together, Misao inadvertently - or not so inadvertently - getting involved in some of Kenshin's troubles. She soon learned that Kenshin is travelling to Kyoto in order to face the madman Shishio Makoto.
Misao, along with the remaining members of the Oniwabanshu at the Aoiya, pledged their support to Kenshin. Okina helped Kenshin find the son of the swordsmith who forged his sakabatou to obtain a new one and helped him locate his teacher, Hiko Seijuro - master of
the Hiten Mitsurugi-ryu.
Misao eventually learned from Okina that Kenshin was once known as the infamous Hitokiri Battousai. Shocked at first, she was not deterred from giving him her support, and she tells him before he leaves the Aoiya that when they first met, she met him as a wanderer, not a manslayer. Her words were reminiscent of what Kaoru had first told Kenshin and they warm his heart once again.
After Kenshin left, Misao soon met Kamiya Kaoru and Myojin Yahiko - Kenshin's friends. She convinced Okina to tell her where he is and she then took Kaoru and Yahiko to Seijuro's home. After a brief reunion, they headed back to the Aoiya and Yahiko suddenly remembered that he forgot to warn Kenshin about Aoshi, who he had seen in Kyoto a few days before. Misao was naturally shocked and demanded to know more about Aoshi. Yahiko reacted defensively, considering that Aoshi and Kenshin had once fought before, but Kaoru complied, grateful for the chance to see Kenshin again.
Misao was shocked to learn that Han'nya and the others had died and she runs back to Aoi-ya for clarification from Okina. She then realized that Okina is not there and Omasu told her that he had gone to fight with Aoshi. She left to go find them immediately but unfortunately, she arrived too late to stop the fight but just in time to see the horrible result of Aoshi's deadly attack on Okina. Aoshi coldly told her to never show her face to him again and to let the Battousai know that he will be waiting for him at Shishio's hideout.
Misao returned to the Aoiya where Okina was treated for his injuries as he remained on the brink of death. Omasu gave her a letter that Okina had written in foresight of the possibility that he would lose the battle with Aoshi and die. In the letter, he told her that she should forget about the Oniwabanshu and go on and lead a normal life as a teenage girl. Headstrong and determined, she crumpled the letter and refused out loud to forget the ninja clan because they were her family and her happiness. She declared that Aoshi was no longer with them and that she will be the new okashira and they would do all in their power to assist Kenshin in his battle with Shishio, even defeating Aoshi himself.
Misao soon proved herself to be a capable, young leader. When a letter from Kenshin arrives detailing Shishio's secret plans to burn Kyoto to the ground, Misao ordered the other Oniwabanshu to inform the people through their messenger pigeons. Prepared in advance, the citizens remained on watch during the night and prevented any of Shishio's men from setting fire to the city.
Kenshin eventually returned to the Aoiya and learned what had transpired since he left. After waking from his coma, Okina asked Kenshin to kill Aoshi. Kenshin, however, promised Okina that he would bring Aoshi back home because he doesn't believe that Aoshi has lost himself completely. Misao, who had been steeling herself to never look up to Aoshi again, was overwhelmed with this knowledge and found herself crying automatically.
While Kenshin goes to fight Shishio, some members of the Juppongatana arrived to attack the Aoiya. Misao assisted Kaoru in fighting the cross-dresser Kamatari but was knocked unconscious from a direct blow to her ribs. Han'nya appeared to her in a dream to tell her that Kenshin had kept his promise and was bringing Aoshi back. She woke up, inspired and motivated once again, and helpeds Kaoru defeat Kamatari.
Misao and the others waited for Kenshin and company to return from the battle and they eventually do, Aoshi included. Misao was overjoyed to see her beloved Aoshi-sama once again, despite what he had done. However, to her disappointment, once he recovered from his injuries, he avoided everyone and did practically nothing but meditate in the temple.
Kenshin remained in Kyoto for about another month, but decided to finally leave with Kaoru and the others. Misao was sad to see them leave but Kenshin cheered her up by assigning her the task of getting Aoshi to smile.
Some time after the end of the battle, Kaoru wrote Misao a letter, asking her to deliver the diary of the late Yukishiro Tomoe - Kenshin's deceased wife whom he had accidentally killed - to Tokyo as soon as possible, before her younger brother Yukishiro Enishi could exact his form of revenge - Jinchu - on Kenshin. However, Misao arrived with the diary too late. Kaoru was dead and Kenshin had fallen into a state of depression. She took the news from Yahiko in stride and snapped him out of his own depression. She told him that it was his duty as Kaoru's student to not give up. She said that she would like to take Enishi's head herself but that would go against Kaoru's Kamiya Kasshin-ryu and Kenshin's vow. Instead, she swore to find Enishi, make him read Tomoe's diary and face the truth then force him down onto his knees and beg for forgiveness before Kaoru's grave. Despite her earlier composure and determination, Misao later cried to herself, mourning the loss of Kaoru. Unbenownst to her, Aoshi heard her.
Misao eventually accompanied Yahiko to the Fallen Village where Kenshin had settled in his despair and attempted to bring him to his senses, but they fail. Meanwhile, Aoshi met with Takani Megumi and began to suspect that Kaoru was not dead. He had her body exhumed and sliced it open to reveal that the body buried was actually an elaborate doll. Ecstatic, Misao and Yahiko began a frantic and rash search to find Kaoru.
With the help of a mysterious old man and resident of the village, Kenshin regained his senses and determination and left the Fallen village and his living hell. Misao joined Kenshin and the others to Enishi's island, where Kaoru was being held captive. With some guidance from Aoshi, she set off the bombs in the water surrounding the island, preventing their small boat from hitting any of them and allowing the group to land on the shore. After Enishi was defeated, Aoshi decided to head back to Kyoto. Misao realizesd Aoshi wanted to leave early because he wanted to rebury Han'nya and the others before winter arrived, supposedly in Kyoto. Misao was happy to go home with "everyone", and Aoshi finally smiled, although Misao didn't see it.
